A Comparative Study of DNA Ploidy Pattern by Flow Cytometry and Ag-NORs in Colon Adenocarcinoma


Abstract
In colon adenocarcinoma, it has been well known that the prognosis of the disease depends on the Duke classification, histologic grade and serum CEA level.
To evaluate the realtionship between conventional method (cellular differntiatio by histologic grade), argyrophilic method(Ag-NORs) and attractive method (flow cytometry), we studied 30 cases of colon carcinoma who were operated at Department of
Surgery, Korean University Hospital, from Mar. 1987 to Feb. 1992.
@ES The results were as follows;
@EN 1. Of the 30 cases of adenocarcinoma, 12 cases (40%) were DNA aneuploid type. Diploid type was 83.3% in well differentiate type, 57,2% in moderate differentiate type and 33.3% in poorly differentiate type. There was a weak positive
correlation
between type of ploidy and cellular differntiations of colon adenocarcinoma (r=0.28).
2. The cytometric S-Phase had a significant difference with cellular differentiations of colon adenocarcinoma (P=0.05) and cytometric G0/G1 Phase and proliferation index had strong correlation with cellular differentiations of colon
adenocarcinoma
(r=0.33, r=0.34).
3. In Ag-NORs study, the mean number of Ag-NORs WAS 5.68¡¾3.77 in aneuploid type and 3.52¡¾1.75 in diploid type. The mean number of Ag-NORs had a significant difference between diploia typs and aneuploid types (P=0.05).
4. There was a weak correlation between mean number of Ag-NORs and cytometric G0/G1 Phase, S-Phase and proliferation index (r=0.28, r=0.26, r=0.28.)
5. The cytometric S-Phase was 8.5¡¾9.0% in diploid type and 19.9¡¾11.7% in aneuploid type, G0/G1 phase was 86.7¡¾9.8% in diploid type and 75.6¡¾11.5% in aneuploid type, proliferation index was 13.3¡¾9.8% in diploid type and 24.4¡¾11.5% in
aneuploid
type. The cytometric S phase, G0/G1 Phase and proliferation index had a significant difference with ploidy pattern (P=0.006, 0.006, 0.007).
